Label-free deep-tissue imaging in vivo using adaptive optical synchronous angular scanning microscopy

Abstract: Light waves propagating in biological tissues undergo wavefront distortion because of spatial heterogeneity of anatomical structure and cellular distribution, etc. This results in blurring of an optical focus and the reduction of the resolving power. We propose label-free adaptive optical coherence microscopy which is free from hardware feedback for corrective updates. Experimental setup is based on wide-field and time-gated interferometric microscopy. Wide-field configuration enables to collect whole optical modes, thereby it offers optimal correction of high-order optical aberration and significant enhancement of point spread function. As a prospective application, we employ it into label-free neuroimaging, which visualizes neural network in a larval zebrafish in vivo.
